So...What is the difference between a regulator and an expansion chamber? Can you use HPA and CO2 in both? Which would be better for a mechanical blowback like a spyder or tppmann?

an expansion chamber is useless with HPA, it just allows the CO2 to expand a little more before reaching the gun itself. if you check in the Ultimate Resource Thread, i think it lists good regs for CO2, because some regs use o-rings and parts that aren't CO2 friendly.

if you have a stock spyder or tippman, a reg isnt going to do you much good, it'll probably just end up restricting air flow and lowering your FPS. to see improvement, you'll need to invest in a high-flowing valve and possibly a new spring kit.

but on the other hand, an expansion chamber is a waste of money as it does nothing for you except give you a good grip on your gun.
